Effects of ethacrynic acid and furosemide on the hormone-mediated adenylate cyclase activation of the hamster kidney.

چکیده
The effects of ethacrynic acid and furosemide on the adenylate cyclase activity of the hamster kidney and heart were examined. Ethacrynic acid and furosemide suppressed the vasopressin-mediated increment of adenylate cyclase activity much stronger than the basal activity of the renal medulla. Ethacrynic acid and furosemide strongly suppressed the parathyroid hormone-mediated increment of adenylate cyclase activity on the renal cortex, though furosemide could not inhibit the basal activity of the enzyme. Ethacrynic acid and furosemide suppressed the 1-adrenaline-mediated increment of adenylate cyclase activity of the cardiac muscle, while furosemide could not inhibit the basal activity. Ethacrynic acid and furosemide suppressed the NaF-mediated increment of adenylate cyclase activity of the renal medulla. Thus, these diuretics seem to inhibit the catalytic unit of the enzyme. Ethacrynic acid and furosemide strongly suppressed the hormone-mediated increment of the adenylate cyclase activity, while Ca** could only depress it to the same degree as it did the basal activity. This difference suggests that these diuretics inhibit not only the catalytic unit of the enzyme, but also other steps of the adenylate cyclase system. Ethacrynic acid and furosemide suppressed the effect of 5'-guanylylimido-diphos-

E VALUATION OF ADENYLATE CYCLASE ACTIVITY IN MITRAL VALVE PROLAPSE
The term mitral valve prolapse (MVP) is used for a particular subset of patients with hyperadrenergic dysautonomia. It occurs when part of a leaflet or both leaflets of the mitral valve extend above the plane of the atrioventricular junction during ventricular systole. The adenylate cyclase activity in MVP dys-autonomia was studied by extraction of enzyme from the erythrocytes from 62 norma...
متن کاملMechanism of resistance to the phosphaturic effect of the parathyroid hormone in the hamster.
The effect of parathyroid hormone and calcitonin on the renal excretion of phosphate, calcium, and cyclic AMP was evaluated in the thyroparathyroidectomized hamster, a mammal apparently reisstant to the phosphaturic effect of parathyroid hormone. Parathyroid hormone did not increase phosphate excretion, although it decreased excretion of calcium and increased urinary excretion of cyclic AMP. Th...
متن کاملAltered Receptors
Receptor disorders may produce specific clinical manifestations resembling hypo-or hyper-function of related endocrine gland. Receptor disorders can be divided into five categories: 1)Altered receptor concentration exists in insulin resistance seen in obsity, familial hypercholesterolemia and in testicular feminization. 2) Altered receptor affinity can be seen in fasted obese patients, acromeg...
متن کاملSpecific binding of (3H) lysine-vasopressin to pig kidney plasma membranes. Relationship of receptor occupancy to adenylate cyclase activation.
A plasma membrane fraction was prepared from pig kidney medulla. It contained lysine-vasopressin-sensitive adenylate cyclase activity (maximum activation: 4to 6fold, apparent Km value for the hormone: about 5 X 1OV M). Lysine-vasopressin was not inactivated by the membrane preparation even at low hormonal concentrations. The specific [3H]lysine-vasopressin (vasopressin) binding sites found are ...
متن کاملThe vasopressin-sensitive adenylate cyclase of the rat kidney. Effect of adrenalectomy and corticosteroids on hormonal receptor-enzyme coupling.
A subcellular fraction prepared from rat kidney medulla contained vasopressin-sensitive adenylate cyclase and vasopressin binding sites. Vasopressin stimulation resulted in an increase in the maximal velocity of the reaction with no change in the apparent K, for ATP. A maximum activation ratio (5 to 10) was obtained at low Mg2+ concentration (0.75 mM) and pH 7.4. The apparent Km for vasopressin...
